This study evaluated the nutrient-diffusing substrate method in laboratory under controlled temperature (20oC), for further application in in situ bioassay enrichment experiments. The experimental unit comprised a 300mL polystyrene pot filled up with 2% agar solution with isolated or combined addition of nitrogen (0.75 M NaNO3) and phosphorus (0.05 M KH2PO4). The topside opening of the pot was covered with plankton net cloth used for periphyton growth. Nutrients daily release rates were determined during 33 days. Diffusion coefficients obtained by regression analysis were similar in all treatments (0.05day-1). Results indicated that nutrient diffusion was not influenced by isolated or combined addition, thus allowing comparison between the treatments. In addition, after 4 days N:P diffusing ratio was kept close to the previously established condition.
